Martina Navratilova defeated Helena Suková in the final, 6–3, 6–2 to win the women's singles tennis title at the 1986 US Open. It was her third US Open title. Navratilova became the first woman in the Open Era to win the title after saving match points en route, doing so against Steffi Graf in the semifinals.

Hana Mandlíková was the defending champion, but lost in the fourth round to Wendy Turnbull.
